DNA extracted from the second cervical vertebra is preferential for STR typing in old human remains

Despite many protocols are proposed for STR analysis of old human remains, the question of preferential DNA source for successful genotyping is still open. Herein we compared STR typing efficiency in DNA samples extracted from different bones: tubular and flat bones, several vertebrae and coccyx from the World War II victim's burials. Partial STR genotypes were obtained from all examined bones but we've got the complete genotypes profile in DNA samples extracted from the specific structure of the 2nd cervical vertebra – odontoid process. Furthermore, a histological analysis of its morphological structure showed some specific features such as distinguishable osteons within Haversian channels and the presence of lamellar structures that can create optimal conditions for DNA preservation in old human remains.
